چکیده

Fourteen different agroresidues were screened for alpha amylase production using Bacillus amyloliquefaciens ATCC 23842. Among them, wheat bran (WB) and groundnut oil cake (GOC) in mass ratio of 1:1 was proved as the best substrate source. Supplementation with 0.01 M KH2PO4 and 1 % soluble starch enhanced the enzyme yield considerably. Maximum enzyme recovery from the solid mass was obtained when extracted with 0.1 M acetate buffer, pH=5.0. Maximum enzyme titer expressed as units per mass of dry substrate obtained was 62 470 U/g after 72 hours of fermentation at 37 °C by using the above solid substrate mixture (5 g) with the initial moisture of 85 % and inoculated with Bacillus amyloliquefaciens of 2·10 CFU/mL.

Bacillus Amyloliquefaciens Alpha-amylase Inhibition by Organic Solvents: A Study on Methanol, Ethanol and Propanol

Alpha-amylase is widely used as an industrial enzyme, and a therapeutic target for which inhibitors are designed. Organic solvents are used to dissolve various compounds that would be studied as moderators of alpha-amylases, but they could themselves affect enzyme activity and stability. Methanol, ethanol and propanol are simple alcohols that may be commonly used to this end, and their effect h...

A statistical approach for the production of thermostable and alklophilic alpha-amylase from Bacillus amyloliquefaciens KCP2 under solid-state fermentation

The bacterial strain producing thermostable, alklophilic alpha-amylase was identified as Bacillus amyloliquefaciens KCP2 using 16S rDNA gene sequencing data (NCBI Accession No: KF112071). Medium components were optimized through the statistical approach for the synthesis of alpha-amylase by the organism under solid-state fermentation using wheat bran as the substrate. The medium components infl...

Comparison of the alpha-amylase of Bacillus subtilis and Bacillus amyloliquefaciens.

The alpha-amylase (alpha-1,4-glucan 4-glucanohydrolase, EC 3.2.1.1) of Bacillus subtilis strain W23 is less negatively-charged than the alpha-amylase of B. amyloliquefaciens strain F, as determined by electrophoretic mobility in polyacrylamide gel at pH 8.6. The alpha-amylase of strain W23 is immunologically unrelated to the alpha-amylase of strain F, as judged by lack of cross-reaction in Ouch...

Production of Alpha Amylase from a Randomly Induced Mutant Strain of Bacillus Amyloliquefaciens and Its Application as a Desizer in Textile Industry

The present study is concerned with the improvement of Bacillus amyloliquefaciens strain UNG-16 for alpha amylase production. The bacterial culture was exposed to UV irradiation at 1.6×10 J/m/S for 15-60 min. However, UV induced viables did not give improved alpha amylase production; therefore chemical mutation using ethyl methane sulphonate (EMS 50-300 μl/ml) was undertaken for 10-60 min. The ...
